RESUMO

Understanding anaerobic biodegradation of ether oxygenates beyond MTBE in groundwater is important, given that it is replaced by ETBE as a gasoline additive in several regions. The lack of studies demonstrating anaerobic biodegradation of ETBE, and its product TBA, reflects the relative resistance of ethers and alcohols with a tertiary carbon atom to enzymatic attack under anoxic conditions. Anaerobic ETBE- or TBA-degrading microorganisms have not been characterized. Only one field study suggested anaerobic ETBE biodegradation. Anaerobic (co)metabolism of ETBE or TBA was reported in anoxic microcosms, indicating their biodegradation potential in anoxic groundwater systems. Non-isotopic methods, such as the detection of contaminant loss, metabolites, or ETBE- and TBA-degrading bacteria are not sufficiently sensitive to track anaerobic biodegradation in situ. Compound- and position-specific stable isotope analysis provides a means to study MTBE biodegradation, but isotopic fractionation of ETBE has only been studied with a few aerobic bacteria (εC -0.7 to -1.7‰, εH -11 to -73‰) and at one anoxic field site (δ2H-ETBE +14‰). Similarly, stable carbon isotope enrichment (δ13C-TBA +6.5‰) indicated TBA biodegradation at an anoxic field site. CSIA and PSIA are promising methods to detect anaerobic ETBE and TBA biodegradation but need to be investigated further to assess their full potential at field scale.

Isotopic techniques have been used to study phenomena in the geological, environmental, and ecological sciences. For example, isotopic values of multiple elements elucidate the pathways energy and nutrients take in the environment. Isoscapes interpolate isotopic values across a geographical surface and are used to study environmental processes in space and time. Thus, isoscapes can reveal ecological shifts at local scales, and show distribution thresholds in the wider environment at the macro-scale. This study demonstrates a further application of isoscapes, using soil isoscapes of 13C/12C and 15N/14N as an environmental baseline, to understand variation in trophic ecology across a population of Eurasian badgers (Meles meles) at a regional scale. The use of soil isoscapes reduced error, and elevated the statistical signal, where aggregated badger hairs were used, and where individuals were identified using genetic microarray analysis. Stable isotope values were affected by land-use type, elevation, and meteorology. Badgers in lowland habitats had diets richer in protein and were adversely affected by poor weather conditions in all land classes. It is concluded that soil isoscapes are an effective way of reducing confounding biases in macroscale, isotopic studies. The method elucidated variation in the trophic and spatial ecology of economically important taxa at a landscape level. These results have implications for the management of badgers and other carnivores with omnivorous tendencies in heterogeneous landscapes.

In this study, the use of co-occurring discriminators of sewage and manure was assessed as a potential way to disentangle sewage and manure sources. A suite of human and veterinary derived chemical markers, which includes pharmaceuticals and compound such as food additives, has been identified for this purpose. The suite was selected in such a manner as to provide additional source characterisation, e.g. differentiating raw versus treated sewage inputs. An SPE-LC-MS/MS method was developed and validated for the determined suite of chemical markers with a detection limit of up to 50 pg L(-1). This represents one of the lowest limits of detection for pharmaceuticals reported in literature. To illustrate the suitability of the proposed method to differentiate sewage and manure inputs to surface water bodies, results from surface water samples collected at monitoring sites corresponding to specific land use types within Ireland are discussed.

Egg traceability/authenticity is a worldwide concern. Stable isotope techniques have been suggested as a tool to address this issue. To further validate the use of these techniques, a research project was undertaken to evaluate what effect sample collection and preparation have on the measured isotopic composition of egg components. The timing of egg collection, the timing of egg preparation after collection, and the use of pasteurisation were investigated. The C, N, O, and S isotopic compositions of egg components from 7 different production systems were measured. Two sets of eggs were collected (4 months apart). It was found that the 'isotopic fingerprint' of a particular production system was maintained over time, and that it may be possible to trace liquid egg products based on isotopic data from fresh eggs. The findings from this study support the integration of stable isotope techniques in egg traceability/authenticity systems.
